Experimental cancer vaccine both treats and prevents brain cancer in mice

Researchers estimate that more than 13,000 Americans received a diagnosis of glioblastoma — a type of brain cancer — in 2022. And additional researchTrusted Source shows the rate of glioblastoma diagnoses is rising.

There is currently no cure for glioblastoma. Treatment options generally include a combination of surgeryTrusted Sourceradiation, and chemotherapy to help slow the growth of tumors.

Now, researchers from Brigham and Women’s Hospital developed a new cell therapy that both helps eliminate current tumors and acts as a cancer vaccine to prevent tumors from reoccurring.

Scientists recently showed promising results testing the new therapy in a mouse model of glioblastoma.

The study was recently published in the journal Science Translational Medicine.

What are cancer vaccines? 

Over the past few years, there has been much research conducted on cancer vaccines.

There are two main types of cancer vaccines currently under research:

  • vaccines preventing cancer from ever occurring
  • vaccines to treat current cancer and stop it from coming back.

Similarly to vaccines administered for the flu or COVID-19, cancer vaccines work to train the body’s immune system to identify and destroy potentially harmful cells.

There are currently two Food and Drug Administration (FDA)-approved cancer-prevention vaccines available:

And so far, the FDA has approved three cancer-treating vaccines:

Brain cancer vaccine

The new study used a mouse model of glioblastoma to test an experimental cell therapy capable of both eliminating current cancer tumor cells and helping to keep tumors from reforming.

“Our team has pursued a simple idea: to take cancer cells and transform them into cancer killers and vaccines,” says Dr. Khalid Shah, director of the Center for Stem Cell and Translational Immunotherapy (CSTI) and the vice chair of research in the Department of Neurosurgery at Brigham and Women’s Hospital and faculty at Harvard Medical School and Harvard Stem Cell Institute (HSCI), and corresponding author of this study.

“Using gene engineeringTrusted Source, we are repurposing cancer cells to develop a therapeutic that kills tumor cells and stimulates the immune system to both destroy primary tumors and prevent cancer,” he explains.

According to researchers, this cell therapy uniquely uses live tumor cells, while most cancer vaccines use inactive tumor cells.

“We engineered the live tumor cells to release dual cell-killing and immunomodulatory agents,” Dr. Shah told Medical News Today. “These engineered therapeutic tumor cells (ThTCs) eliminated established glioblastoma tumors in mice by inducing cell killing and activating antitumor immune cell trafficking.”

“This efficacy of ThTCs translated into a survival benefit and long-term immunity in primary, recurrent, and metastatic cancer models in immunocompetent and humanized mice. The incorporation of a double kill-switch in ThTCs ensured the safety of our approach. Therefore arming naturally neoantigen-rich tumor cells with bifunctional therapeutics represents a promising cell-based immunotherapy for solid tumors and establishes a roadmap towards clinical translation.”

And, he said, unlike inactivated tumor cells, living tumor cells possess a unique potential to home to and target tumors.

“Therefore, engineering tumor cells to express therapeutic agents is a rational approach that takes advantage of their natural source of neoantigensTrusted Source,” Dr. Shah explained. “Our tumor cell-based bifunctional therapeutic strategy by transforming living tumor cells into potent agents that concomitantly drive direct tumor killing and antitumor immunity.”

Next-generation vaccine

Dr. Shah stated this therapy could potentially be used to treat other cancers other than glioblastoma. And he said we may see a cancer vaccine like this available for public use in three to five years.

“We are developing [the] next generation of autologous and allogeneic engineered tumor cell-based vaccines and are hopeful that our therapeutic strategy will have the potential to impact patients by preventing tumor progression, recurrence, and metastasis,” he added.

MNT also spoke with Dr. Santosh Kesari, a neuro-oncologist and director of neuro-oncology at Providence Saint John’s Health Center and chair of the Department of Translational Neurosciences and Neurotherapeutics at Saint John’s Cancer Institute in Santa Monica, CA, and Regional Medical Director for the Research Clinical Institute of Providence Southern California, about this study.

He said that while immunotherapyTrusted Source is an exciting approach for cancer that has changed the outlook for many cancers such as melanomaTrusted Sourcelung cancerTrusted Source, and gastrointestinal cancerTrusted Source, scientists have not made much progress yet in using immunotherapies for brain cancers.

“New approaches are needed and certainly this new study expands our understanding of how to modify and use the native tumor cells to potentially generate a more robust immune response in vivo,” Dr. Kesari explained.

“The live cancer cells mentioned in this article are engineered to directly kill tumor cells and activate the immune system and reduce tumor immunosuppression. So this system allows both activation of [the] immune system to recognize and kill cancer, and at the same time also get rid of the brakes that prevent the activated immune system from doing its job,” he noted.

As for the next steps in this research, Dr. Kesari stated much work needs to be done to translate the preclinical work in this paper into a human proof-of-concept.

“[I] would like to see more animal studies looking at combining with other immunotherapies such as checkpoint inhibitorsTrusted Source, and how this could also work in combination with radiation and chemotherapy,” he continued. “There are significant hurdles and costs to be dealt with in making live cell products but can be overcome with time and resources.”

Apple Fitness+ unveils new offerings for the new year

Beginning January 9, the service will introduce a new Kickboxing workout type, a brand-new sleep theme for meditations, a Beyoncé Artist Spotlight, new Time to Walk guests, and three new trainers

Apple Fitness+, the award-winning fitness and wellness service that is welcoming to all, is introducing new ways for users to improve their overall well-being in the new year. Starting Monday, January 9, the service will launch Kickboxing, a new total-body cardio workout type. A brand-new meditation theme, Sleep, will join the nine other themes in the Meditation library, all designed to make users’ practice more impactful than ever before. To help them get started, a new program called Introduction to Meditations for Sleep — designed to help users wind down before bed and drift off to sleep — will launch.

Additionally, Artist Spotlight will launch new workouts featuring music by Beyoncé, including songs from her latest album, RENAISSANCE, bringing her iconic music to the service. Time to Walk will kick off its fifth season with new guests, including Golden Globe-nominated actor Jamie Lee Curtis, late-night talk show host Amber Ruffin, Olympic champion figure skater Nathan Chen, and German actor Nina Hoss. Fitness+ will also launch two new Collections — 6 Weeks to Restart Your Fitness and Level Up Your Core Training — as well as welcome three new trainers to the team.

Kickboxing and New Trainers

Kickboxing workouts can help build full-body fitness, and are a great way to build strength, stamina, coordination, and balance. Starting January 9, Fitness+ will add Kickboxing as a new total-body cardio workout type. Each workout will consist of a distinct round of moves followed by one final round, which will combine the moves users just learned into an all-out one-minute interval. No equipment is required, and workouts will be 10, 20, or 30 minutes long.

Kickboxing will be led by two Fitness+ trainers. Jamie-Ray Hartshorne, who users know and love from Treadmill, HIIT, and Time to Run workouts, trained in muay thai and professionally competed in Thailand.

Additionally, Nez Dally will join the trainer team to lead Kickboxing workouts. The muay thai fighter made history when she became the first woman to compete in Thailand wearing a hijab. In her workouts, she brings the drive of a high-achieving athlete to empower users with every jab, cross, and kick.

In addition to Dally, two more new trainers will join the Fitness+ trainer team in January: Brian Cochrane for HIIT and Jenn Lau for Strength. Cochrane, who hails from Scotland, grew up playing a variety of sports, making his HIIT training a versatile practice. As a former singer in an indie band, he continues to be inspired by the way music and movement come together.

Lau co-founded a gym in her hometown of Toronto, and she is just as passionate about inclusivity as she is about helping people stay active. She brings tenacity and playfulness to every lift and lunge she teaches.

Introducing Sleep Meditation

Meditation on Fitness+ is designed to help users release tension and anxiety. A brand-new Meditation theme, Sleep, will be added to the Meditation library, joining the nine other themes, including Calm, Gratitude, Resilience, and Creativity. The robust library is designed to make the practice of meditation more powerful and useful than ever before. New sleep meditations will be added every week, and each practice can be done as part of a wind-down routine or while drifting off.

To help users get started with sleep meditations, a new program, Introduction to Meditations for Sleep, will launch. The program consists of four 20-minute meditations that conclude with five minutes of relaxing music. The Artist Spotlight series — which dedicates an entire workout playlist to a single artist — expands with new workouts featuring music by Beyoncé, including songs from her latest album, RENAISSANCE, bringing her iconic music to the service. On Monday, January 9, seven new workouts featuring the singer’s music will be available across Cycling, Dance, HIIT, Pilates, Strength, Treadmill, and Yoga. The energy and themes in each workout are inspired by the album’s notion of rediscovering what makes individuals unique and powerful to help uplift users in the new year.

Fitness+ will also roll out two additional Artist Spotlight offerings: the Foo Fighters on Monday, January 16, and Bad Bunny on Monday, January 23.

ADNOC Allocates 15 Billion to Low-Carbon Solutions

Abu Dhabi, UAE – January 5, 2023: ADNOC, a reliable and responsible provider of lower-carbon intensity energy, today announced a bold new strategy to progress the world-scale decarbonization of its operations. 

The announcement follows the guidance by ADNOC’s Board of Directors in November 2022 to accelerate delivery of its low-carbon growth strategy and the approval of its Net Zero by 2050 ambition. This builds on ADNOC’s strong track record as a leading lower-carbon intensity energy producer, which includes its use of zero carbon grid power, a commitment to zero flaring as part of routine operations and deployment of the region’s first carbon capture project at-scale.

Acting on the Board’s guidance, ADNOC has allocated $15 billion (AED55 billion) to advance an array of projects across its diversified value chain by 2030. These projects will include investments in clean power, carbon capture and storage (CCS), further electrification of its operations, energy efficiency and new measures to build on ADNOC’s long-standing policy of zero routine gas flaring. ADNOC will apply a rigorous commerical and sustainability assessment to ensure that each project delivers lasting tangible impact. 

Throughout 2023, a suite of new projects and initiatives will be announced, including a first-of-its-kind CCS project, innovative carbon removal technologies, investment in new, cleaner energy solutions and strengthening of international partnerships. Together with the recent formation of the ADNOC’s new Low Carbon Solutions and International Growth Directorate, these represent tangible and concrete action as the company reduces its carbon intensity by 25% by 2030 and moves towards its Net Zero by 2050 ambition.

Building on ADNOC’s Al Reyadah facility, which has the capacity to capture up to 800,000 tons of CO2 per year, the company will announce plans to deploy technologies to capture, store and absorb CO2 by leveraging the UAE’s geological properties while preparing for its next major investment to capture emissions from its Habshan gas processing facility. Combined with ADNOC’s planned expansion of its carbon capture capacity to 5 million tons per annum (mtpa) by 2030, the UAE will be firmly established as a worldwide hub for carbon capture expertise and innovation. 

ADNOC’s expansion of CCS is planned to support the significant scale-up of hydrogen and lower-carbon ammonia production capabilities in Abu Dhabi as ADNOC advances a world-scale 1 million tons per annum (mtpa) blue ammonia production facility at TA’ZIZ, the industrial services and logistics ecosystem that is enabling the expansion of the Al Ruways Industrial City, as well as Abu Dhabi’s wider chemicals, manufacturing and industrial sectors. To-date, ADNOC has already delivered test cargoes of low-carbon ammonia to Europe and Asia. 

ADNOC’s expansion of its new energy portfolio will largely be delivered through its stake in Masdar, the UAE’s clean energy powerhouse with over 20 gigawatts (GW) of clean energy today and plans to increase its capacity to 100 GW by 2030. Masdar is also spearheading the UAE’s drive to develop a leading position in green hydrogen.  

Since January 2022, ADNOC has received 100% of its grid power supply from Emirates Water and Electricity Company’s (EWEC) nuclear and solar energy sources, making it the first major company in the industry to decarbonize its power at scale through a clean power agreement of this kind. ADNOC also concluded a $3.8 billion deal to build a first-of-its-kind, sub-sea transmission network in the MENA region, connecting ADNOC’s offshore operations to the onshore power network, with the potential to reduce ADNOC’s offshore carbon footprint by up to 50%.

Building on the multi-billion capital investment in decarbonization projects, ADNOC is working closely with its international partners and stakeholders across the energy value chain to collaborate on technology, best practices and policy to support and drive global decarbonization efforts.

Amiri Unveils a Pop-Up At the Mall of the Emirates

Los Angeles-based brand Amiri has returned to Dubai for its second pop-up store in the Mall of the Emirates. The brand brings its wavy west coast aesthetic with its latest Pre-Spring22 menswear collection. The collection includes a range of big fits: from hand-painted hoodies and leather biker jackets to simple logo T-shirts and ripped jeans. Either way, the collection personifies Amiri’s L.A. roots and affinity for classic all-American staples. 

The pop-up itself is minimal yet styled with exacting detail: marble and wood hybrid plinths, a Jean Royere-inspired white couch that has been seen all over Instagram and sought-after Piere Jeanneret wood and cane chairs.

Founder and creative director Mike Amiri started his journey in fashion with hand-embellished denim creations. Eventually, the designer grew the brand into a global behemoth. In recent months, the brand has rapidly expanded, opening swanky new stores in Las Vegas, New York City and London. Yet, the majority of the brand’s collections are locally created in Los Angeles at the brand’s in-house studio.

Microsoft and Viasat announce new partnership to deliver internet access to underserved communities globally

New collaboration through the Microsoft Airband Initiative will increase internet availability for 5 million people across Africa and an additional 5 million people around the world by the end of 2025

REDMOND, Wash. — Dec. 14, 2022 — On Wednesday, Microsoft Corp. (NASDAQ: MSFT) and Viasat (NASDAQ: VSAT) announced a new partnership to help deliver internet access to 10 million people around the globe, including 5 million across Africa. Viasat, a global communications company, is the first satellite partner to work with Microsoft’s Airband Initiative, and together they will deepen Airband’s work in the Democratic Republic of the Congo, Nigeria, Guatemala, Mexico, and the United States, as well as prioritize expanding the program to Egypt, Senegal and Angola to deliver much-needed internet connection, often for the first time.

This first of its kind global partnership for Airband is an important step in reaching the Initiative’s expanded goal of delivering internet access to a quarter of a billion people across the world, including 100 million people on the continent of Africa, by the end of 2025.

According to the International Telecommunication Union at the UN, roughly one third of the world’s population — or 2.7 billion people — have still never used the internet. Satellite allows internet access to reach remote areas that previously have had few, if any, options for conventional connectivity. Working together, the companies will combine expertise and assets to help enable telehealth, distance learning and education, precision agriculture, clean power, and other services to reach new areas through the transformational provision of power and connectivity. The companies will collaborate to provide and pilot technologies including, but not limited to, satellites (both geostationary orbit and low earth orbit) and fixed wireless.

“We believe access to the internet is a fundamental right and that digital skills create and enable economic prosperity for people, businesses and governments. Through our Airband Initiative we will extend high-speed internet access to 100 million people on the continent of Africa and to a quarter of a billion people living in unserved and underserved areas across the world by 2025,” said Teresa Hutson, Microsoft’s vice president of Technology and Corporate Responsibility. “Working with Viasat, we will use satellite to reach remote areas that previously have had few, if any, options for conventional connectivity. Together, we will be able to rapidly scale and expand Airband’s reach, exploring a wider pipeline of projects and new countries where we haven’t yet worked.”

Nearly one third of the world’s population is lacking online access to education, better medical care, business opportunities, connection with family and more. And most of this population lives in just 20 countries across Africa and the Global South. Universal, affordable internet access is part of the United Nations’ Sustainable Development Goals (SDGs), and by focusing a large portion of this new partnership on Africa, Microsoft and Viasat are working to deliver connectivity and digital literacy for better education, healthcare and economic opportunity in critical markets.

The family friendly development, which builds on the strengths of The Sustainable City – Dubai, is a walkable community in the surrounds of open green spaces, leisurely walkways, and community farming plots.

The project is underpinned by a central green spine that runs the length of the community, featuring parks, lakes, and biodomes where vegetables will be grown and distributed throughout the community. With a strong focus on enabling an active community, The Sustainably City – Yas Island will also be home to a range of communal facilities and amenities, including an equestrian centre with stables, a track and two arenas, a gym and multiple pools, cycling and jogging tracks, football, basketball and padel courts, retail and F&B outlets, a nursery, a centre for autism, and a green and sustainable mosque.

Dubai records over AED10 bn in weeklong real estate transactions

DUBAI, 6th January, 2023 (WAM) — A total of 2,173 real estate transactions worth over AED 10 billion were conducted during the week ending 6 January 2023, according to figures released by the Dubai Land Department (DLD).
A total of 270 plots were sold for AED 2.25 billion, 1,903 apartments and villas were sold for AED 4.89 billion.
The top three transactions were a land in Marsa Dubai sold for AED 623.07 million, followed by a land that was sold for AED 77.23 million in Al Thanyah Fifth, and a land sold for AED 623.07 million in Marsa Dubai in third place.
Al Hebiah Fifth recorded the most transactions for this week by 133 sales transactions worth AED 386.38 million, followed by Jabal Ali First with 49 sales transactions worth AED 228.15 million, and Al Merkadh with 12 sales transactions worth AED 125 million in third place.
The top three transfers for apartments and villas were an apartment was sold for AED 119 million in Island 2, a was second in the list sold for AED 115 million in Island 2, and thirdly it was an apartment sold for AED 75 million in Island 2.
The sum of the amount of mortgaged properties for the week was 1.85 billion, with the highest being a building in Burj Khalifa, mortgaged for AED 300 million.
101 properties were granted between first-degree relatives worth AED 1 billion.
